I have been involved in various research themes involving the human enteric parasite Entamoeba histolytica, particularly on the cell biology and evolution of mitosomes, a class of mitochondrion-related organelle present in this parasite. My previous research theme was focused on the characterization of novel mitosomal β-barrel proteins in E. histolytica. Its major contribution was the discovery of MBOMP30, a novel, Entamoeba-specific, mitosomal outer membrane β-barrel protein. I was also part of collaborations that aimed to better understand the mechanisms involving mitosome fission and protein targeting. Currently, we are collaborating to characterize lineage-specific mitosomal membrane proteins. This endeavor is crucial because membrane proteins are known to play various essential roles in protein and metabolite transport, lipid exchange, organelle dynamics and biogenesis. Recently, a strong trend has emerged towards the investigation of membrane proteins that mediate membrane contact sites, enabling inter- or intra-organelle crosstalk. There is a possibility that our candidate proteins may be lineage-specific receptors, channels, enzymes, or components of novel complexes. Thus, our findings could answer or support new hypotheses on the lack of detectable homologs of key components (e.g. channel protein on the outer membrane, protein import machinery on the inner membrane) in the E. histolytica mitosome. The knowledge generated could support the development of new parasite-specific drugs, as the proteins of interest are Entamoeba-specific. Lastly, I am also involved in the lipidomic profiling and preliminary investigation of fatty acid elongation in E. histolytica as well as the purification and proteomic analysis of E. histolytica exosomes. |
